Inbred strains of rats differ in their susceptibility to both spontaneous and chemically-induced mammary carcinogenesis. We have been using genetic breeding strategies between resistant and susceptible rat strains, followed by linkage analysis to identify the mammary carcinoma susceptibility (Mcs) gene(s) responsible for the mammary carcinoma resistance trait in the inbred Copenhagen (Cop) rat. Simple sequence repeat (SSR) markers from the literature as well as from our chromosome-specific libraries were used to map the Cop rat genome and fine-map the regions involved in the resistance phenotype. So far we have found 4 loci (3 resistance and 1 enhancer) that show linkage with the mammary carcinoma resistance phenotype. The locus on chromosome 2 (termed Mcs1) is believed to contain the major tumor resistance gene, while other loci such as Mcs2 on chromosome 7, Mcs3 on chromosome 1, and Mcs4 on chromosome 8, represent relatively weaker genes.
